| In recent years, the incidence of cancer increased persistently. Malignant tumor hasbecome one of the main factors that affect human health, so it is very urgent to treattumor. But the treatment to tumor today, such as chemotherapy, radio therapy, madeserious damage to the normal tissue which provided the limitation of application. Inother aspects, the small molecular targeted anti-cancer showed apparent irreversibletolerance. Therefore, we need to explore a new kind of drug which has high efficiency,high specificity and low toxicity.With the deepening research on cancer, more and more research showed that theimportant role of glycosaminoglycans(GAGs) in the process of proliferation anddevelopment of tumor progress. So this article explore the change of the structure andcomposition of glycosaminoglycans after the compound effecting cancer cells, andthis would be significant to find an anti-cancer drugs.In this article,5-hydroxy-6,7,8,4’,5’-five methoxy flavone compound (referredto as5HPMF) is a kind of extraction and separation of traditional Chinese medicine(TCM) Tangerine Peel. Previous studies have found that it has the effect of inhibitingcancer cell proliferation. This article mainly studies the influence of the structure andcomposition of glycosaminoglycans from the cancer cells which treated with5HPMF.The experiment mainly measured the concentration of GAGs samples with carbazolemethod, the quantity of FGF/GAG/FGFR trimer with flow cytometry, theconcentration of monosaccharide of GAGs samples with HPLC. This article detectedthe changes of concentration, quantity and structure of GAGs samples treated with5HPMF in order to find the mechanism of5HPMF’s anti-cancer effects.This article first linked the inhibition of GAGs synthesis and the inhibition ofcancer cell proliferation to study the effect of flavonoids on cancer cells. This articleinnovated a method to extract GAGs samples from cancer cells which treated with5HPMF. Experiments show that this method was stable and especially suitable for tumor cells which has small amount of glycosaminoglycans. In addition, this methodhas not been reported yet.This article detected the concentration of GAGs samples by carbazole method andthe concentration of glucosamine and galactosamine by HPLC method repectively.The study found that the concentration of GAGs samples is low, but the differencebetween the control group and5HPMF treated group is not obvious. The HPLCanalysis data showed that the relative content of heparan sulfateglycosaminoglycans and chondroitin sulfate glycosaminoglycans is not significantlyaffected by5HPMF. on the other hand, this article detected the different quantity ofFGF/GAG/FGFR trimer by flow cytometry and this method has not been reported.The results showed that the peak of FGF1/GAG/FGFR1C,FGF9/GAG/FGFR3C,FGF2/GAG/FGFR1C and almost had no shift,while the results showed that the peakof FGF7/GAG/FGFR2B, FGF8/GAG/FGFR3C trimer shifted when treated with5HPMF. It has been reported that the specificity structure of GAGs is essencial for theform of the FGF/GAG/FGFR trimer. So this data showed that the fine structure ofGAGs is effected by5HPMF.This article combined the aboved analysis results of glycosaminoglycans withintracellular signal transduction pathway of protein which has the regulated function.Based on the systematic analysis, this article could explain the molecular mechanismsthat5HPMF influences cancer cells.The article focus on the mechanism of5HPMF’s anticancer effects which influenceglycosaminoglycans. This research also played an important role in the furtherdevelopment of dried tangerine or orange peel and the advancement of TCM(Traditional Chinese Medicine) modernization. |
