Objective: Studies showed that caffeine increases thermogenesis and fat oxidation,suppresses fat accumulation and body weight gain. Further, Astilbin increases lipoproteinlipase activity and inhibits the rate-limiting enzyme activity of cholesterol synthesis,regulate lipid metabolism. But the combinational effect of caffeine and astilbin on lipidmetabolism is unclear. Therefore, the present study investigates the effects of caffeine andastilbin on lipid metabolism and its molecular mechanism in high-fat diet-induced obesemice.Methods:80female ICR mice were randomly divided into8groups: normal group (I),high fat diet group (II),0.4%astilbin (III),0.06%caffeine (IV),0.03%caffeine+0.2%astilbin (V).,0.03%caffeine+0.4%astilbin (VI),0.06%caffeine+0.2%astilbin (VII),0.06%caffeine+0.4%astilbin (VIII) for8weeks respectively, body weight was weightedevery week. Organs and intraperitineal adipose tissues (IPAT) were weighted at the end ofexperiment. Serum lipids concentrations and liver lipids levels were measured. Theactivities of lipid metabolism related enzyme, the mRNA and protein expression of lipidmetabolism related enzyme were determined in liver.Results:(1) Compared with II, the body weight of V, VI, VII, VIII groups and the IPAT weightof VII, VIII groups were significantly reduced.(2) Compared with II, TC level of VII, VIII groups, TG level of VI, VII, VIII groupswere significantly decreased in the serum and liver.(3) Caffeine and astilbin combination inhibited FASN and HMG-COAR activities,and increased CAT andAMPK activity in liver.(4) Caffeine and astilbin combination reduced the mRNA expression of FASN, HMG-COAR, SREBP1c, and enhancedAMPK, HSL,ATGL, CAT mRNAexpression in liver.(5) Caffeine and astilbin combination inhibited the protein expression of FASN,enhance AMPK, HSL, ATGL protein expression, and increased the phosphorylation ofHSLprotein in liver.Conclusion: The results showed that affeine and astilbin regulated the mRNA andprotein expression of lipid metabolism related enzyme, inhibited FASN, HMG-COARactivity, increase CAT, AMPK activity, inhibited fat synthesis, promoted fat oxidation, andreduced the lipids level in serum and liver. It causes the suppressive effect on fataccumulation and body weight gain. Moreover, it was indicated that caffeine and astilbinwas synergistic in anti-obesity effect.
