Antibiotics are the chemicals which can interfere with functional phylogeny of the otherliving cells. They are the activity of pathogens or other type of secondary metabolitesproduced by the microbes (including fungi、bacteria、actinomycetes) or higher animals andplants in the life. The antibiotics have been used extensively in clinic treatment and animalfeedings. The ecology was damaged and human health was threatened by the antibioticresidues. Liquid-liquid extraction and solid-phase extraction have been most commonly usedto pretreatment for determining antibiotics. However, these methods are still immature formmost analyses due to consumption of time and a large amount of volatile, toxic, pollutionorganic solvents. Additionally, the methods can cause secondary pollution, high cost and awaste of time.The goal of the project was to develop a novel and non-toxic method for the separation/enrichment of antibiotic in water. The new method is based on the solvent sublation and ionicliquid-salt aqueous two-phase system combined with ultraviolet spectroscopy. The primaryresearch contents and conclusion are as follows:(1)The flotation systems of [Bmim]BF4/NaH2PO4and [Bmim]Cl/K2HPO4wereestablished.(2)The new separation/concentration method of doxycycline hydrochloride andsulfadiazine was put forward depending on [Bmim]BF4and NaH2PO4. Moreover, antibioticscould be both measured and analyzed by UV spectrophotometry. The last, ionic liquid-saltaqueous two-phase extraction experiments of the same chemicals have been done in the sameconditions.(3)A separation/concentration method of sulfadiazine and doxycycline hydrochloridewas studied depending on [Bmim]Cl and K2HPO4.Similarly, antibiotics could also bemeasured and analyzed by UV spectrophotometry. And ionic liquid-salt aqueous two-phaseextraction experiments of the same chemicals have been done.(4)A separation/concentration method of dopamine hydrochloride was further discussedbased on [Bmim]BF4and NaH2PO4. The dopamine hydrochloride could be determined andanalyzed by high performance liquid chromatography. We found out the optionalexperimental conditions through orthogonal test. Ionic liquid-salt aqueous two-phaseextraction experiments of the same chemicals have been done in the optional conditions. Theconclusion of tests was further proved. From all the results, we know that the efficiency of doxycycline hydrochloride based on[Bmim]BF4and NaH2PO4is the highest. Under the same conditions, the separating efficiencyof ionic liquid-salt aqueous two-phase flotation is higher than extraction in all respects.In all, the experiment of flotation/separation antibiotics in water by ionic liquids aqueoustwo-phase system flotation was based on [Bmim]BF4and NaH2PO4.It may be regarded as acombined method of flotation and solvent extraction with the advantages of both techniques.This new technology has many virtues, such as non pollution, nontoxic, high enrichmentfactor, high efficiency, negligible phenomenon etc. The method is suitable for determining thetrace antibiotics of environmental sample.
