Loquat, originated from China, is the ideal fruit. Not only is it juicy, delicious andfull of nutrition, but also it wins medical scientists’ favor for having experienced fourseasons. In addition to eating the flesh, people also often process the flesh intopreserved fruit and brew fruit wine or vinegar and so on. But the Loquat nuclearalways is regarded as the waste to be abandoned, which, consequently, lead to thesource-consuming. Recently, it was reported that the extracts from the nuclear areimmune to the mice. However research about the composition rarely was studied.In this essay, under the condition of having not been fully utilized and developed,the extraction and analytical method was established to determine the functionalingredients in the Loquat nuclear, which provided theoretical basis on making full useof Loquat and reference to exploiting the nuclear1. The extraction condition of pentacyclic triterpenoids in Loquat nuclear hasstudied. Through wavelength scanning of the extracting solutions of four agents(ethanol, methanol, ethyl acetate and chloroform), the ethanol was determined to bethe best solvent. By the single factor and orthogonal experiments L9(43) tests, whenextracting temperature was95℃, ethanol concentration was80%, time and thematerial-liquid ratio were60min,1∶30respectively, the yield of triterpenes wasmaximal. And after range and SPSS analysis, the order of each factor was:temperature>concentration>temperature>material-liquid ratio>time. Andtemperature, ethanol concentration and the ratio were significant to the experimentresult.2. Using colorimetric method to determine the total content of triterpenes. Theresults showed:when λ was544nm, perchloric acid and5%Vanillin-glacial aceticacid were1.00mL and0.30mL respectively, the total content of triterpenes was3.455mg/g with RSD1.08%. And different areas contains various amount oftriterpenes. Then optimizing the conditions of measuring ursolic acid by HPLC. Theextracting conditions were identical with the triterpenes’ with different factor order.When λ was210nm and the ratio of methanol to water was95∶5, the content of ursolic acid was1.5889mg/g. The two detective methods both displayed goodreproducibility and reliable recovery with ranging from89%to104%.3. The methods of extracting and testing polyphenols from Loquat nuclear werestudied. Firstly, through the orthogonal experiments L9(33) based on every singlefactor, the color reaction of Folin-Ciocalteu with polyphenols was optimized. Thatwas10%sodium carbonateon1.5mL,20%Folin-Phenol3.5mL, reaction time70min.Then by the single factor and orthogonal experiments L9(43) tests, the extractioncondition was established: temperature80℃, concentration of ethanol50%, time120min, the ratio of sample to solvent1∶20. Temperature and ethanol concentrationwere significant to the result. Meanwhile after repetitive tests and recovery tests,when n=5, the content of polyphenol in the seed was2.368mg/g with the averagerecovery rate97.9%, and RSD was1.34%. So the method established to analyze thepolyphenol in the seed was feasible and simple with good stability and accuracy.
