Biodiesel is renewable clean bioenergy as it can be produced from vegetable oils,animal fats and microbial oil. Microbial oil has many other oils incomparableadvantages, thus has great development potential. Using microorganism such as algaeand yeast, ferment cheap raw material to produce oil and preparation into biodiesel hasbeen widely studied.In this paper, using Trichosporon cutaneum as oil production strain and theinulin as carbon source to produce oil and preparate biodiesel. Methods of oilextraction and biodiesel preparation has optimized, the oil composition and thephysical and chemical properties of biodiesel has tested. The results were as follows:(1) Ultrasonic assisted the acid-heating method was selected as the way to oilextraction, which used the single factor method, the Plackett-Burman experimentaldesign, the steepest ascent and the reponse surface analysis to optimize. The bestconditions of Trichosporon cutaneum oil extraction were as follows: acid treatmenttime27.3min, ultrasonic time7.5min, rate of extracting solution (VCHCl3:VCH3CH)1.7:1. On this condition, the oil yield forecast of4.57±0.01g/L. The validation test foroil yield was4.54±0.01g/L.(2) Under the effect of ultrasonic, Trichosporon cutaneum oil and methanolmade esterification reaction with KOH as catalyst. The single factor method, thePlackett-Burman experimental design and the reponse surface analysis were used tooptimize it. The best conditions of ultrasonic auxiliary alkali catalysis methodpreparation biodiesel were as follows: molar ratio of methanol to oil7.8:1, ultrasonicpower333W, catalyst dosage1.24:1. On this condition, the biodiesel yield forecast of94.01±1.34%. The validation test for biodiesel yield was94.01±1.34%.(3) Gas chromatography was used to determine compositions and content of thebiodiesel, results showed that methyl oleate content was the largest. Via detected, the physical and chemical properties of biodiesel were in line with the BD100standard inour country.
