However, the presence of trehalose synthasehydrolysates unconverted maltose, due to its chemical structure and high similarity oftrehalose, a conventional method of separating difficult, high production costs, butalso affect the purity of the trehalose. Therefore, a cost-effective research solution toremove residual maltose hydrolysis method is necessary.In this study, a composite mutagenesis breeding osmophilic high conversion rateof maltose yeast strains used in the mutant strain was purified trehalose process highpurity trehalose. The method as a new method for purifying enzymatic production oftrehalose, trehalose simplify the purification process and reduce production costs. Theconclusions of this study are as follows:1. A higher conversion rate of maltose Saccharomyces cerevisiae (Saccharomycescerevisiae, Angel) as the starting strain, the use of ultraviolet mutagenesis withNTG mutagenesis methods maltose strain high conversion rate, get three good mutantstrains, namely G-4B4, E-14A5, G-4C3, three strains of mutant strain fermentedtrehalose synthase enzyme solution after48h, maltose conversion rates were85.03%,86.03%and87.37%, compared with the original strain increased by4.7times.2. In the mutant yeast strain G-4C3as the research object, under certain yeastextract concentration1.8g/L, pH5.5conditions by single factor test and responsesurface of the medium test strains were optimized. Determined by response surfaceoptimization test each medium concentration of metal ions in potassium dihydrogenphosphate1.07g/L, magnesium sulfate1.87g/L, ferric sulfate0.14g/L.3. G-4C3mutant analog trehalose synthase enzyme solution at28℃, underfermentation conditions120rpm after48h, a final concentration of96.69%trehalose,trehalose purified strains good effect.
