Innate immune plays a very important role in organism defense mechanism, because it is the first line of defense against microbial invasion from outside in multicellular organism. Compared with mammals, the innate immune system of Drosophila does not have similar t-lymphocytes,b-lymphocytes and acquired immune system which can specifically identify pathogenic microorganisms. But it can provide quick and efficient response to microbial infection, which proves Drosophila has a excellent natural immune system. The present research results show that Drosophila has three main ways of natural immunity:the first one is the humoral immune which against pathogenic microorganisms though antimicrobial peptide produced by fat body; The second is the cellular immune which relys on cell phagocytosis and packages; The last one is melanization which works though producing melanin to deposite on the wound and kill the pathogenic microorganisms.Protein Rgn(regeneration) is one of the members of the C-type lectin superfamily, and it includes a signal sequence and a C-type Ca2+-dependent lectin domain(CTLD).It is coded by gene rgn. The research show that it has a very important role in individual development,tissue regeneration and wound healing. It participates in all stages of growth and development of Drosophila, and it is one of an important protein in Drosophila growth. Thus speculated it may also participate in the Drosophila innate immune response. Experiments show that the ability of resistance to Candida albicans significantly decreased in rgn deficit type breed and the number of blood cells significantly improve compared with Wild-type breed (w1118). The phenomenon of pupal death and abnormal development of vein were also observed. This suggests that lacking protein Rgn can cause the immune system dysfunction of organism. At the same time, the experiment detects the phagocytosis of cells, the ductility of the cell membrane actin and the ability of trigger melanotic encapsulation by wound. There is no significant change comparing with Wild-type breed.In addition, preparation protein Rgn polyclonal antibody makes the research of protein level come true. It is able to reveal the cellular localization and expression more directly and accurately and provide the experimental basis for the research of protein Rgn.
