Histone deacetylases (HDACs) catalyze histone deacetylation, regulate the status of histone acetylation, and play an important role in the modification of chromatin structure and the regulation of gene transcription. To study the function of HDA902 gene, the encoded sequence of HDA902 gene was cloned by PCR method, the encoded protein sequence was analyzed by bioinformatics method, and the GFP-HDA902 fusion gene expression vector was constructed and expressed in the onion epidermal cells to determine the subcellular localization of HDA902. Results:(1) The ORF of HDA902 gene was 1500 bp, encoding a protein containing 499 amino acids. (2) HDA902 protein shared a conserved domain (HDAC3) with the HDACs in other plants. HDA902 protein was relatively close to HDAC (XP 007200978.1) from Prunus persica and HDA19-like (XP 008348955.1) from Malus domestica in evolution. (3) The promoter sequence of HDA902 gene contained multiple stress-related and light-responsive cis-acting elements, including 5UTR Py-rich stretch, ARE, ACE and HSE. (4) HDA902 protein mainly localized in the cytoplasm. (5) HDA902 gene in Populus trichocarpa was expressed in leaf, stem and root. Its expression level in the stem and leaf was higher than that in the root. The expression of HDA902 gene was in response to abiotic stresses. Its expression level was down-regulated by salt stress, but up-regulated by cold stress. (6) To study the functions of HDA902 in stress responses, a plant expression vector pROK â…¡-HDA902 was constructed and transformed into Nicotiana tabacum by Agrobacterium-mediated method. The expression of HDA902 in tobacco improved the cold tolerance of transgenic plants.
