SbbHLHl doesn’t interacting with a bHLH domain protein of Sorghum bicolor (L.) Moench(SB02G040140) in our Yeast two-hybrid experiment.This suggests that SbbHLHl maybe not a party ofBMW (bHLH-MYB-WD40) tripolymer, but constitude dipolymer with itself or other bHLHdomainprotein, take part inlignin transcript regulationnetwork.Using SELEX technology we isolated the DNA sequence that combining with SbbHLH1.Thesequences contain E-box motif(CANNTG). In order to find the envidence of interaction between SbbHLHl and the promoters of genes in lignin biosynthesis pathway in Arabidopsis thaliana, we conducted combine experiment between SbbHLHl and the sequence of these genes’promoter. The result indicates that SbbHLHl can combine with the promoter ofAtPAL. This suggests that SbbHLH1 can regulate lignin biosythesis in Arabidopsis thaliana.Agrobacterium tumefaciens with pSTART-SbLIMl were used to infect of Populus.The positive transgenic populous plants were obtained by kanamycin selection and PCR,The lignin content of SbLIM1were decreased.The 5 plants of SbbHLH1transgenic populous we got were all chimeras.The lignin content and QT-PCR results of some positive balso show that the expression of some lignin biosythesisgenesis reduced.These suggest that SbbHLHl and SbLIM1transgene Populus has the potential to be biomass energy source.
