Chloroplast (cp), a crucial semi-autonomous organelle, is now becoming more and more important in the plant genome research. Sapindus mukorossi and Tetratigma hemsleyanum Diels et Gilg, as two medical plant, have vital economic and medical value. As little cpDNA research has been done with these two medical plant, we published the whole cp genome of these two medical plant and did some related studies:(1) We did comparative studies of the extraction methods of cp DNA. High salt method was found the best through weight, conditions of chloroplast and the assembly result.(2) The complete cp genome of Sapindus mukorossi is 160,481 bp and the complete cp genome of Tetratigma hemsleyanum Diels et Gilg is 159,889 bp. The GC content and the repetitive sequences have been detected for both plants.(3) Comprehensive analysis has been done with these two plant:the rps19 gene of IRa/LSC was lost in the 3 plants of Vitales and the ycfl gene of SSC/IRa was also lost in Tetratigma hemsleyanum Diels et Gilg. For all 6 plants of Spindales and Vitales, the coding region is more conserved than the non-coding region, and the LSC and SSC regions are more divergent than the two IR regions.(4) 5 data sets (rbcL, atpB, matK, rbcL+atpB+matK and 53 protein coding regions) have been used to reconstructe the phylogenetic relationship of 41 eurosids plants. The result of the data set of 53 protein coding regions was more accuracy with extremely high supporting rate.
