Ethanol inducible expression system(ALCA) is a class of chemically-induced promoter well-studied in recent years, which can achieve rapid and efficient induction of expression of the target gene. FT(Flower Locus T) proved to be a integration factor is the part of the flowering network, which can effectively promote flowering. GmFT2 a is one of 10 GmFT homologous genes in soybean.Over expression GmFT2 a, could promote early flowering. The soybean crop is a typical short day、strong regional,、single species and adapt to a narrow range,so the regulation of GmFT2 a is of great significance to soybean breeding. photoperiodic response and mechanism of flowering and maturity of molecular design breeding.To artificially control the temporal and spatial expression of GmFT2 a, achieve the early promotion of soybean flowering,we build transformed vector contains ethanol inducible expression system driven reporter gene vector(ALCA-mini35S: GUS) to transformed soybean hairy root.By the relative expression assay of GUS and GUS activity, evaluate the efficiency of induction in soybean hairy roots. On the basis of GUS reporter gene transformation,build the inducible system driven GmFT2 a vector and transform hairy root,get ALCA-mini35S:GmFT2a positive hairy root and then measure the expression of GmFT2 a. The results showed that when added ethanol by root-inducing, GUS expression and GUS activity of ALCA-mini35S: GUS positive hairy root was significantly increased,and when add the 0.05% ethanol,the mean GUS expression and GUS activity were higher than other groups.Ethanol expression system was established in soybean hairy roots. When positive ALCA-mini35S: GmFT2 a soybean hairy root was trated by 0.05% ethanol, GmFT2 a expression was significantly increased. Then builded transformed vector contains ethanol inducible expression system driven reporter gene vector(ALCA-mini35S: GmFT2a) to transformed Arabidopsis, after ethanol spraying, the transgenic plants’ days of breeding to flowering and total leaves of were less than wild types’, show early flowering phenotype. In this study, we get the realization of the GmFT2 a expression.This study show that ethanol inducible expression system can function in soybean hairy root,by means of ethanol inducible expression system,can achieve the expression and regulation of the GmFT2a.
