Takydromus amurensis belongs to the reptilia, Squamata, Lacertiformes, Lacertidae and this species mainly lives in Changbai mountains and their extension area of eastern northeast of China. This research mainly includes three parts: the complete mitochondrial genomes sequencing of T. amurensis, the studies on the phylogeny of Lacertidae in China and the cytochrome B gene of different geographical populations of T. amurensis.The main results are as follows:1.The complete mitochondrial genome of T. amurensis consists of 17333 bp and including 13 protein coding genes, 22 tRNA genes, 2 rRNA genes and a non-coding control region(D-loop). The mitochondrial genome sequence is similar to the structure of vertebrate mitochondrial genomes have been reported. The base content that A is 31.23%, C is 26.06%, G is 13.91%, T is 28.8%. The content of A + T(60.03%) is greater than the content of G + C(39.97%).The GC deflection value is 0.304 while the AT deflection value is 0.040 in this species. The complete mitochondrial genome sequence of three species(T. amurensis,T. wolteri and T. Sexlineatus) were compared and analyzed, the results show that genetic relationship between T. amurensis and T. wolteri nearly.2. We do phylogenetic studies on the mitochondrial genomes of 12 species of 4 genera of lizards in China with the whole length, sequence structure, the content of the bases and the genetic distance. The results show that all the mitochondrial genome sequence length is 17046~19914 bp, the average length is 18471 bp. It consisted by 13 protein gene, 22 tRNA genes, 2 rRNA genes and 1 control region(D-loop). The content of G is the lowest in the four kinds of bases, the average is 13.46%, the content of T and C are closed, which accounted for 28.99% and 26.56% respectively, the A content is the highest and the average is 30.99%. The content of A + T(59.98%) is greater than the content of G + C(40.02%). The genetic distance of four genus showed that Platyplacopus and Takydromus recently, their genetic distance was 0.207. Eremicas and Takydromus of the genus genetic distance, as far as 0.321.3. The length of Cytb gene in five different geographical populations of T. amurensis is 1143 bp. A total of 13 haplotypes are defined in 27 individual and 27 variable sits are identified. The Haploype diversity(Hd) is 0.932±0.025 and Nucleotide diversity is 0.00487±0.00038. The estimates of Tajima ’s D and Fu’ sFS show that the five populations relative to neutral evolution of species diversity and no significant deviation(P > 0.1).
