One strain M136 producing maltogenase was isolated. Separation of maltooligosaccharide with macroporous resin was explored. With maltoligosaccharides as the substrate, the measurement method of the enzyme activity was established. The culture condition for maltogenase production was optimized and maltogenase was characterized.1. The researches began with a static absorption and a dynamic test on macroporous resin to absorb and purify the maltooligosaacharides, to find out the largest volume of the sample and the corresponding volume of detergent. Then the effect on macroporous resin’s absorbability of such factors as the solution density of the sample and current speed were studied. The results showed that the optimum conditions were as follows:the current rate was 1.0 mL/min and the density of sample was 50 mg/mL.2. The optimal enzyme production conditions for strains:with maltooligosaccharide as the carbon sources, carbon source concentration for 50 mg/mL, yeast powder as a source of nitrogen, leaching nitrogen source concentration for 2 mg/mL, fermentation time for 24 h, temperature, medium for 37℃ initial pH 6.5.3. Ammonium sulfate saturation between 40% and 70% sediment contains most get the starch sugar degrading enzymes vigor.The optimum temperature for enzyme reaction was 45℃. The Maltogenase exhibited maximum activity at pH6.5, and was stable at pH6-7.5. The effect of metal ion on the Maltogenase activity was studied. The enzyme activity was aetivatied by K+, Mg2+and Mn2+. The enzyme activity was significantly inhibited by Fe3+ and Ca2+. Cu2+, Zn2+ and Cr3+completely inhibits the M136 Maltogenase activity.
