Rhodobacter sphaeroides 2.4.1 has been widely studied in photosynthesis, nitrogen fixation and membrane protein. YidC is Oxal/Alb3/YidC family protein, it could assist the insertion, assembly and folding of the other membrane proteins, and play a significant role in the energy metabolism in E. coli, but there was no any report on the study of the structure and function of the YidC of the Rhodobacter sphaeroides 2.4.1.We got the 1753 gene mutant RSRif (1753-) by inserted a fructose-induced yidC gene and a kanamycin resistance gene into the 1753 gene of the R. sphaeroides 2.4.1, but failed to get the chromosome yidC gene knock out mutant of the RSRif (1753-).So the function of the yidC gene was studied by the complementation of the expressed YidC of R. sphaeroides 2.4.1 in the E. coli YidC-deletion mutant FTL10. We found that the FTL10 containing the expression plasmid of the yidC gene of the R. sphaeroides 2.4.1 could grow without arabinose, indicating that YidC of R. sphaeroides 2.4.1 has the similar function as that of the E. coli. We also found that C-terminal last 5 amino acide residues of YidC of R. sphaeroides 2.4.1 is very important for the proper fuction of the YidC of R. sphaeroides 2.4.1 in the E. coli, because this 5 amino acid residues deletion YidC mutant could not complement the FTL10.As a membrane protein, it’s not easy to get high yield recombinated YidC protein. To get high yield of YidC, we tried different expression vectors and finally got the recombinated YidC with expression vector pIND4 in R. sphaeroides 2.4.1 and E. coli with higher yield in R. sphaeroides 2.4.1 than in E. coli.
