Cytokinesis is the last stage of mitosis, in which chromosomes and cytoplasm are averagely distributed into two daughter cells if it is normally completed. The defect of cytokinesis will lead to the formation of multiple nucleus cells, which means the aneuploidy of chromosomes, for example, chromosomes are always aneuploid in many kinds of cancer. The meaning of research on cytokinesis is to understand the mechanism of the formation of some cancer and try to find out the specific way to cure cancer.The most studied on cytokinesis is the affection of MAP (microtubule associate protein), for example, PRC1 (protein regulating cytokinesis 1) and Mklp1 (motor-kinesin-like protein 1). When the stability and dynamic of the complex of MAPs become abnormal, the defect of cytokinesis come into being. However, the research on actin associate protein is focused on the formation and contractility of contractile ring, while we know little about the actin on telophase especially on abscission. It is still unclear that how GTPases and their GAP change their activity and affect the cell during this period.Arf6 is ADP-ribosylation factor 6, a GTPase, which participates the remodeling of actin skeleton on cell migration. One of its GAPs (GTPase activating protein), ACAP4/ASAP3, is the main object in this dissertation. It is a multiple-domain protein that the most two important domains are Arf GAP domain and PH domain, the former is to activate Arf 6 and the latter is to bind to PI(4,5)P2 and be activated. Some research on Arf6 suggests that Arf6 plays a role in linking microtubules to membrane near midbody and maintaining the stability of midbody. So it worths to study how the GAP ACAP4 works in cytokinesis.In this thesis, I studied the influence of the knockdown of ACAP4 on cytokinesis, and compared the affection of the wild-type and modification-mutants of ACAP4 on cell skeleton during cytokinesis. I had some conclusion and may it helps the following research.
