Objective: In order to explore rare actinomycetes diversity from the mangroves in of Fujian province and to isolate new actinomycetes for discovering compounds of pharmaceutical importance.To lay the foundation for future resource development.Methods:We collected 26 soil samples from Fujian Mangrove rhizosphere and 14 kinds of selective media were used to isolate actinomycetes. The isolates were identified by morphology and 16 S r RNA gene sequence analysis.We use pure culture method to screen type strain.After the production of actinomycetes microsections,we use optical microscopy(OM) and scanning electron microscopy(SEM) to observe their apparent morphological characteristics.After the work that actinomycetes selected were preliminary identified is completed,we filter and discard a lot micromonospora by useing specific primer M558 F and C1028 R. The PCR reaction is completed by the use of bacterial universal primers 27 f and 1492 r.136 strains of actinomycetes were identified by phylogenetic analysis based on 16 S r RNA gene sequences.Screening of biosynthetic genes based on PCR was used to detect potential producer strains of non-ribosomal peptide synthetaseand and halometabolites.Results:In total 803 strains of actinomycetes were isolated from the 26 soil samples by the plate dilution method.After dereplication and removing the similar strains based on colony-characteristics, of them 136 strains were selected and then characterized by phylogenetic analysis based on sequences of 16 S r RNA gene. 16 S r RNA gene sequences phylogenetic analysis showed that these strains belonged to 7 families including 13 genera. 40.4 % of the 136 strains were identified containing NRPS gene fragment and 13.2% identified containing halogenase gene fragment.Conclusion: This study shows that the mangroves in Fujian are rich source of rare actinomycetes and micromonospora is the dominant populations.The actinomycetes from mangroves environment have the potential of producing non-ribosomal peptide synthetaseand and halometabolites.
