With the development of research on epigenetics, people gradually realize the importance of mi RNA. mi RNA participates in a variety of complex regulatory pathways, and has important implications for key events and the occurrence of a variety of diseases in the development. In this study, mi R-1904 is located on mouse chromosome 13. And it is in intron 8 of Pde4 d. Translation products of Pde4 d gene can take part in c AMP/CREB and c AMP/EPAC signaling pathway and produce functions through hydrolyzing second-messenger cyclic adenosine monophosphate( c AMP), having huge effect on animal health. Previous studies stated that, intronic mi RNA probably produce synergistic or antagonistic effect on the expression of the hosted gene.There are many researches concerning Pde4 d, which is the hosted gene of mi R-1904. However,there lacks studies about mi R-1904. Thus, this study does research on the expression profile of mi R-1904 in mouse embryo stage, to investigate the temporal and spatial patterns of mi R-1904 expression in mouse embryos. Then, in order to determine the effect of mi R-1904 on embryo development, through vivo experiments on pregnant rat tail vein injection of mi R-1904 lentiviral vector, we test the indicators of pregnant rat and sub embryo. Lastly, in the livo experiment, the specific expression of mi R-1904 was selected as the model, and the relationship between Pde4 d and mi R-1904 was preliminarily discussed.Mi R-1904 in mouse embryo stage exists specific pattern of expression, mainly in all kinds of neural tissue and some specific organs. Most of the nervous tissue( brain, spinal cord, dorsal root ganglia) and the olfactory cortex during the entire period have maintained a high level of expression. In addition,lungs and kidneys also remain relatively high levels of expression. However, liv er, heart and other organs of mi R-1904 in expression levels fluctuate over time. In vivo experiments, after the injection of mi R-1904 carring slow virus, the body weight of pregnant rats was lower than normal level and the coefficient of lung organs is larger. At the same time, it can induce the abnormal of blood circulation related organs(heart, l ungs, liver and kidney) in pregnant rats. The abnormal rate of offspring embryos increases, Sub embryo minfies, delaying development, especially, the abnormal situation of lung and kidney is more prominent. The expression of mi R-1904 in the lung and kidney of the sub embryos increase significantly. This phenomenon demonstrates with the help of the blood circulation system of pregnant rat, mi R-1904 lentiviral vector can act on sub embryos through blood tire barrier. Mi R-1904 was preliminary proofed to target Pde4 d by the software prediction and luciferase reporter gene technique report experiments. The specific expression of mi R-1904 in lung and kidney in sub embryos as model, this study found overexpressed mi R-1904 can not affect Pde4 d at the level of transcription. But the phenomenon of decrease of Pde4 d protein in lungs explains overexpressed mi R-1904 can act as negative regulators of Pde4 d on protein level.
