| Simuliidae is a kind of important zoonoses vector insects, because of its blood-sucking habit and it also spread disease, as many countries accached great importance to the ecological habit and prevention of it. But not all the blackflies are vector insects, so the accurate identification of blackfly became the basis of the work. Black flies are a challenge for traditional taxonomy because of their small size and structural homogeneity because of the shape similarity, especially within in the same group of relatives, although cytogenetic method can identify to species, but it requires specific development phase of the sample and the operation is time-consu ming. So it is necessery to use molecular methods and other comprehensive measures to study the molecular taxonomy and phylogenetic analysis, and it also can lay the foundation for the reaearch of the prevention and control of blackfly.In this paper, the specimen were identificated according to morphological classification method what were collected and analysis from seven signs in jiangxi province, we get 4 morphospecies:Simulium (Nevermannia) aureohirtumnr, Simulium (.Simulium) quinquestriatum, Simulium (Simulium) bidentatum, Simulium (Simulium) reptans, others specimens were identificated to subgenus, Extract mitochondrial COI gene from the specimen which have been identificated by morphology, then analysis the component, system development and genetic difference by MEGA 5.0, the result is consistent with the morphological identification results. At the same time, we sequece and analysis the entire mitochondrial genome sequences of S.aureohirtumnr by step method. The result shows that the arrangement of S.aureohirtumnr’s mitochondrial genome genes is consistent with the imaginary insects ancestor’s:trnI-trnQ-trnM. |
