Agar is the water-soluble polysaccharide with extracted from Gelidium amansii.The main ingredients are agarose and agaropectin.It has great application prospects in food,medicine,clinic and biochemistry area.In view of its gelling properties,it is widespread use of laboratory cluture.Agarase can hydrolyze polysaccharide and it is a kind of seaweed tool enzyme,meanwhile has important value in single cell separation and enzymatic degraded cell wall into making protoplasm.Also,agarase in molecular biology there are also many aspects of applications.Studies of agarase-producing strains have theoretical significance and application value.From dalian coastal waters Gelidium screened relative higher enzyme activity of agarase-producing strains.Through the morphological characteristics,physiological and biochemical characteristics study its classification status.Vibrio agarivorans-2 prefer 28℃,pH from 7 to 8,NaCl from 2% to 4%,The strain is facultative anaerobes.By the test of single factor and RSM(Response Surface Methodology),the optimum ferment conditions an agar-degrading marine bacterium Vibrio agarivorans-2 were determined as follows:culture time 48 h,culture temperature 28 ℃,initial pH 7.5,inoculum 8%,volume 40 m L/100 mL,shaking speed 160 r/min,and minimize the passage times.From RSM text the best conditions is established of culture time 46.8 h,initial pH 7.4,shaking speed 167 r/min.The enzymatic properties of agarase was preliminary studied.Vibrio agarivorans-2activity was stable in the pH range of 7-8 and under 50℃.
