In the chapter one，The content related tumors were introduced, including thereasons for its formation, human hazards, as well as its raw tumor markers andexplained in detail, and describes the various methods used for chemical analysis,such as chemiluminescence and fluorescence method, a brief description of its basicprinciples and points out which of these methods commonly used in the detection ofthe current summary of our detection levels were to some extent, and futureprospects were briefly discussed.In the chapter two，we designed conformation-switching aptamer molecule thatcould be circularized without ligation DNA. As compared to traditionalmethods in which the elongated product from RCA could result in high signals, inthe present study the petite PPi molecule was converted to ATP with adenosine5’-phosphosulfate (APS) and ATP sulfurylase to result in higher signals. Based onthe proposed method, ATP can be detected at1.0×10-10M. Meanwhile, the methodhas significant implications for real applications, such as detections of ATP in Ramoscell and in human serum.In the chapter three, chapter collar designed molecular beacon containing afluorophore and quencher (MB5), in the DNA enzyme, the solution cyclizationoccurs under the action of shear enzymes, fluorophores separating the quencher, the release of a fluorescent signal. Under optimum conditions, can be detected in1.0×10-8M ATP, and8.0×10-9M GSH. And recovery in K562cells was alsosatisfactory.
