KCTD10(potassium channel tetramerisation domain-containing10) gene belongs to the PDIP1gene family, which contains a BTB/POZ domain, a potassium channel tetramerisation (K-tetra) domain (a relative of BTB/POZ domain) at the N-terminus,and a PCNA-binding motif at the C-terminus. KCTD10can be induced by TNF-a,and interact with PCNA and the small subunit of DNA polymerase δ.The full-length of zebrafish KCTD10cDNA is3716bp,in which the open reading frame encodes a313amino acid residues peptide. The KCTD10gene is highly conserved during evolution, and encodes a highly conserved protein in the human, mouse,zebrafish. In order to further study the fuction of KCTD10in animal development,we use the zebrafish as a model to explore the spatial and temporal expression patterns of KCTD10in zebrafish and synthetise a specific KCTD10Morpholino to inhibit KCTD10gene expression in the development of the zebrafish. We here show that transcripts of KCTD10were expressed in zebrafish embryos at all developmental stages and in different organs, and strong expression of KCTD10was found in brain, heart, liver, gills, fins and reproductive cells,while knockdown of KCTD10in embryos resulted in pericardial edema in the heart, hypoplasia of the brain, weak blood circulation, blood retention,heartbeat slow down and defects in the trunk structure. In addition, these phenotypes were associated with abnormal expression of three heart-chamber markers including cmlc2, amhc and vmhc, and abnormal expression of vascular marker genes flk-1. These data suggest that kctd10plays an important role in the development of heart and skeletal muscle. While the embroys injected with the same dose of control MO show no obvious difference in the developmental pattern,compared with widetype embryos.These results suggest that zebrafish KCTD10gene have an important role in initiating embryonic development and subsequent formation of tissues and organs such as brain, heart and blood vessels.
