URO gene encodes a C2H2Zinc-Finger protein and belongs to a plant-specific gene family. It is the excessive expression of URO gene that leads to the increasement of free auxin’s content in uro mutant. Currently, URO gene is the only reported transcription factor which regulates the auxin homeostatic. The loss of function mutant of the gene had not been found yet. Therefore, a detailed analysis of URO gene’s expression pattern is particularly important for exploring the function of URO gene and revealing the mechanism of auxin metabolism in plant.In previous research, the upstream promoter of4.4Kb,2.2Kb and1.1Kb were cloned and fused with the reporter gene GUS. In this study, the expression pattern of URO gene was achieved through GUS signals analasis and situ hybridization. Our results show that expression of URO gene had distinct specificity of time and spaces. The expression pattern of URO gene was quite similar to the distribution of auxin signal. It’s possible that URO gene could regulate Arabidopsis vegetative growth and reproductive growth through auxin concentration regulation. In addition, part of the coding region of URO gene was involved in it’s expression regulation.The GH3gene at5g13380and at4g18780, a cellulose synthase gene, were cloned and over expressed in Arabidopsis in this work. That will be benefit for further related studies of URO’s downstream genes.
