In this study, we first screened, purified a neurotoxin(named Anntoxin)as research samples from the cDNA library of the skin of the the tree frog Hyla annectans. Mainly from the analgesic and anti-inflammatory effects of animal models for the study of target, to detect the main function of Anntoxin.Anntoxin has60amino acid residues, and it is a kind of tetrodotoxin-sensitive voltage gate control to sodium ion channel inhibitor. Sodium channel has been shown to be a target for the treatment of pain. We use several neuropathic pain in animal models and persistent inflammatory pain (tail-flick test, acetic acid-induced writhing test, formalin-induced paw licking, carrageenan-induced paw edema) to detect the reorganization Anntoxin analgesic and anti-inflammatory role. In all models, the r-Anntoxin shows a strong analgesic activity, while also significantly inhibited the secretion of tumor necrosis factor-a and cyclooxygenase-2. Histopathological studies have shown, the r-Anntoxin inhibition of the carrageenan-induced subcutaneous edema. All the results indicated the r-Anntoxin having a strong analgesic and anti-inflammatory activity.Because skin is the most important interface between the toad internal milieu and the outer environment, and acts as the first barrier against microorganism, highly expressed Anntoxin in the skin may furnish skin with a strong chemical weapon to fulfill its functions.
