In recent years,hermatypic corals bleaching phenomenon was increasingly seriousand appear degradation of ecological communities among world scale due to climatechange and human activities. In this study,we used mutation sites and sequencepairwise differences to find the most suitable molecular marker for the analysis ofgenetic diversity in Porites lutea population. We discussed the genetic structure andgenetic diversity of P. lutea population among four geographic regions including totaleleven groups and evaluated genetic differentiation of different groups andconnectivity among groups using the molecular marker we chosed. This study canprovide genetic basis for protection and restoration of damaged coral reefs. The mainresults are as follows:1. ITS gene was a better marker than COI gene to analyse genetics of P. luteapopulation by comparing sequence mutation loci and sequence differences. The mostsuitable molecular marker to analyse genetic structure and genetic diversity of P. luteapopulation was ITS1+ITS2.2. We chose four geographic regions as Taiwan,Daya bay,Sanya and Indonesia Lembeh Strait with a total of11groups270coral individuals in this study. Except for Daya bay region populations,the population genetic diversity in the rest three regions were all abundant,there were low genetic differentiationand hight gene exchange in local small groups of each geographic population,the whole group experienced population expansion. Daya bay population turned into degraded ecological communities due to a variety of reasons,the genetic diversity in local coral colony was low,but the high gene exchange value implied there were no geographic isolation among local groups and the local groups can be used to repair local coral communities with each other.3. The gene flow analysis between China and Indonesia reef buiding coral groupsshowed a value which was greater than4, this value can validate the kinship indirectlybetween Chinese costal regions and Xisha islands and the islands of the South China Sea through molecular biology method.
