Objective: To investigate the effects of hepatic vagotomy on Gadd45 g and Nox4 expression after partial hepatectomy in rats, and then to explore the effects and mechanism of vagus nerve in liver regeneration.Methods: 108 male SD rats were divided into 2 groups randomly: partially hepatectomy(PH) and partially hepatectomy after hepatic vagotomy(PHV). Rats in both groups were killed at 0h, 2h, 12 h, 24 h, 36 h, 48 h, 72 h, 120 h and 168 h after operation respectively. Immunohistochemistry method and Western Blot method were used to detect the expression of Gadd45 g and Nox4 of regenerating liver tissues respectively. Real-time PCR was used to detect the expression of Gadd45 g mRNA and Nox4 mRNA of regenerating liver tissues.Results:(1) Immunohistochemistry method and Western Blot method revealed that the expression of Gadd45 g in PHV group rats was decreased at 12h~48h after operation compared with PH group and achieved the lowest amount at 48 h. Then it was increased and reached peak at 72 h. The expression was decreased at 72h~120h and increased at 120h~168h. The expression of Gadd45 g in PHV group rats was significantly lower than PH group rats at 2h, 12 h, 24 h, 120 h and 168h(P<0.05).(2) RT-PCR revealed that the expression of Gadd45 g mRNA in PHV group rats was increased at 2h and decreased at 2h~24h after operation compared with PH group. It remained low level at 24h~120h and increased greatly at 168 h. The expression of Gadd45 g mRNA in PHV group rats was significantly lower than PH group rats at 24 h, 72 h, 120 h and 168h(P<0.05).(3) Immunohistochemistry method and Western Blot method revealed that the expression of Nox4 in PHV group rats was increased at 2h~12h and decreased at 24 h after operation compared with PH group. Then it fluctuated. The expression of Nox4 in PHV group rats was significantly lower than PH group rats at 2h, 24 h, 48 h, 72 h, 120 h, 168h(P<0.05).(4) RT-PCR revealed that the expression of Nox4 mRNA in PHV group rats was decreased at 2h~12h and keeped low at 12h~48h after operation compared with PH group. Then it increased at 72h~168h. The expression of Nox4 mRNA in PHV group rats was lower than PH group rats and had significant difference at 2h, 12 h, 48 h, 72 h, 120 h after operation(P<0.05).Conclusion:(1) Hepatic vagotomy may has a role in promoting liver regeneration.(2) The regulate mechanism of hepatic vagotomy to rat liver regeneration may by inhibiting the expression of Gadd45 g and Nox4.
