Transgenic technology was a biotechnology which developed in the 1980 s and people began to produce transgenic animals with superior traits under this background. Transgenic animals were applied to the livestock, the pharmaceutical industry, life sciences, food science and other fields and it makes transgenic animals research flourish, showing a good prospect and was received the wide spread attention.Since the past two decades,there has been a rapid development on transgenic animals research and a variety of transgenic animals can be accessed.However, There were many problems to be solved for achieving commercialization in the transgenic livestock breeding, such as,the low resistance of transgenic animal disease, the high mortality, the poor performance and the abnormal physiological functions. These issues not only affects the healthy of transgenic animals,but also produces a potential threat for the human health, the social stability and the environmental safety.In order to study the impact of exogenous gene against the host immune function, we choosed F1 generation transgenic mice Myo G and Myf6 as the research object and study the effects of exogenous gene on the immunity of cellular, humoral and the m RNA expression levels of immune cytokine. It promotes further the application of transgenic technology in genetic breeding.The main contents of this paper are as follows:1) The genome of F1 generation transgenic mouse were extracted and were identified with ordinary PCR method to filtering mouse with foreign gene.The results showed that six transgenic Myo G gene mouse and seven transgenic Myf6 gene mouse were received.So these mouse with exogenous gene can be used for the following study.2)The copy number of F1 mouse was detected by absolute quantitative PCR method.The results showed that the number of transgenic Myo G gene mouse was roughly 5-7 copies and the number of transgenic Myf6 mouse was one or two copies.3)The spleen index, the lymphocyte proliferation, the apoptosis rate of lymphocytes and the number of T lymphocyte on the transgenic mice were evaluated.The results showed that the spleen index,the lymphocyte proliferation were significantly decreased in transgenic mice;T lymphocytes of transgenic Myf6 gene mouse were significantly lower(P <0.01),and transgenic mouse with Myo G gene also declined, but the difference was not significant individually;The apoptosis rate of lymphocyte on transgenic mouse increased significantly compared with the negative control group(P < 0.01).That showed that the immune function of transgenic mouse is suppressed.4)The m RNA expression of IL-2,IFN-γ, IL-4,IL-6 and TNF-a of transgenic mouse spleen was tested by fluorescence quantitative PCR.The IL-2,IFN-γ m RNA expression on Myo G mouse and Myf6 mouse were decline in different degrees compared with control group and it indicated that the cellular immune of transgenic mouse was inhibited;The expression of IL-4 m RNA in Myf6 mouse turn significantly decreased(P<0.01),it proved that the humoral immune function of Myo G mouse was normal and the humoral immune of Myf6 was still restrained; The expression of IL-6 and TNF-a on some transgenic mouse slightly increased,suggesting that the immunity of transgenic body has declined and the inflammation has occurred.5)The relationship of between exogenous gene copy number and the immunity of two cattle gene transfer mouse were compared and we found that the number of Myo G mouse were significantly more than Myf6 mouse.But the spleen index, the proliferation of T,B lymphocytes and the number of T lymphocytes of Myo G mouse were significantly higher than Myf6 mouse and it proved that the immunity of transgenic mouse decreased was not caused by the copy number.
