In order to investigate community structure of invertebrate resting eggs in the Yangtze Estuary, this research was performed from the cycle estuary, permanent estuary and small river estuary.DNA barcode COI sequence, based on identification of invertebrate resting eggs, provides an experimentally accurate and efficient method. This study used single egg DNA barcoding to set up DNA barcode library, and was the first time to combine the resting eggs’ size, morphology, library and the species length measuring instrument. We had identified 52 kinds of invertebrate resting eggs, including 14 species of cladocera、11 species of rotifera、4 species of copepod and another 23 kinds of invertebrates.Whether the Yangtze Estuary had invertebrate resting egg bank or not could be explained based on the Chongming and Hengsha East Shoal and the results firstly revealed periodic water’s invertebrate resting egg bank structure. The multiple stations of Chongming and Hengsha East Shoal resting eggs’ vertical distribution trend generally increased first and then decreased, with the peak appearing in 4cm-6cm below the surface. By studying 4 loci of the permanent water Qingcaosha Reservoir, we found the A1 and A5 loci’s vertical distribution trend generally increased first and then decreased, with the surface seeing more distribution than the bottom, the B1 and B5 loci’s vertical distribution density trend generally increased first and then decreased, the surface and deep resting eggs’density were higher than the middle, species and density of resting eggs difference inside and outside, that reflected human activities played an important role for egg bank structure; Through studying Binjiang Park, Shangyi Lake and Yingtao river’s 10cm sediments of the resting eggs of community structure, we found the Yingtao river’s species more richer than Binjiang Park and Shangyi Lake, that reflected the diversity of invertebrates in the flowing water was higher than the closed type.By improving the method of HotSHOT, we achieved a rapid and accurate identification of single resting egg. Our results were collected as the table of invertebrate resting eggs’ types and measurements in parts of sediments in the Yangtze Estuary, which can be used in the illustrated handbook concerning with species of invertebrate resting eggs. Based on many previous researches about zooplankton egg bank, our study widened the objection scopes by investigating invertebrate resting egg bank, which extended the domain of eggs’ research. Moreover, it was the first time to study resting eggs’ community in cycled estuary in Yangtze Estuary and we have proved that abundant eggs could be collected in Chongming and Hengsha East Shoal, which provided research basis of regulation on invertebrates’ population and community by resting eggs in Yangtze Estuary.
