Flowering is controlled by diverse enviromental cues, and its molecular mechnisms on transcriptional, translational and epigenetic levels have been widely studied. Nuclear pore complex (NPC) is the key bridge of substance communication between nucleus and cytoplasm. However, the function of nucearporins on flowering and circadian clock regulation remains uncovered.We bought 22 mutants of eight nucleoporins in Nup107-160 sub-complex from Arabidopsis Biological Resource Center (ABRC), and identified 17 homozygous mutants.nup160-1/-2/-3 mutants show significantly flowering early besides diverse developmental phenotypes, regardless of environmental photoperiodic and temperature conditions. However, the nup160 mutants flower earlier in long day conditions than in short day conditions, indicating the nup160 mutants remain the ability to response to light photoperiod. The resluts suggest that Nup160, as a nagetive regulator, plays a role in both the photoperiod pathway and the thermosensory pathway to manipulate flowering time of Arabidopsis.In addition, we carried out the leaf movement assay to measure circadian period of nup160 mutants, and found that the period of nup160 mutants (32 hours) longer than that of wide-type (26 hours). Obviously, Nup160 anticipate in the regulation of circadian period.To explore the possible molecular mechanism of Nup160 to regulate flowering and circadian clock and its interaction with other nucleoporins in Nup107-160 sub-complex, we constructed a series of vectors for the following study, including overexpression, GFP-labled, and promoter analysis.The results provide reliable evidence and substantial materials for elucidating the molecular mechanism of Nup160 gene to regulate flowering and circadian clock, and also lay the foundation for further exploring other functions of nucleporins in Nup107-160.
