Potato (Solanum tuberosum L.) is a kind of crop which widely grown in theworld. Potato starch is widely applied in the industrial field, through the regulation ofplant starch synthase or expression of heterologous enzymes in vivo to alter thecomposition and structure of starch. New physical and chemical properties of starchwhich is manufactured can bring enormous economic and environmental benefits.Higher plants FtsZ1genes control chloroplast division. On account of amyloplast andchloroplasts are derived from plasmids, so FtsZ1gene can also regulate theamyloplasts division. Through the transgenic technology to inhibit FtsZ1geneexpression and control amyloplasts division, accordingly, affecting starch properties.In application of transgenic technology, building a complete regeneration system andan efficient transformation system, to obtain the transgenic plants is the preconditionwhich used transgenic technology to study the FtsZ1gene expression influence ofstarch properties.In this study, take four different genetic backgrounds of potato cultivars as thematerials, studying the regeneration system of four varieties of potato, screening andoptimizing the regeneration system, in order to set up a corresponding regenerationsystem. Through Agrobacterium tumefaciens strains GV3103mediated antisenseFtsZ1genes into different potato varieties to analyze the feasibility whatAgrobacterium GV3103is used for potato transformation and obtain thecorresponding efficient transformation system. At the same time, to study geneexpression of FtsZ1transgenic plants. The results were as following:(1) The "Favorita" and the "Atlantic" are suitable to use in stem section forexplant callus regeneration system.(2) The "Gannongshu No.2","Shepody" and the "Atlantic" are suitable forusing microtuber discs for explant regeneration system of directdifferentiation.(3) Antibiotics are Cef for Agrobacterium strain GV3103mediated genetictransformation of potato and the best bacteriostasis concentration is200 mg/L.(4) In the Agrobacterium strain GV3103mediated genetic transformationsystem, the "Gannongshu No.2" and "Shepody" are suitable for usingmicrotuber discs for receptor material conversion system,"Favorita" issuitable for the stem segments as receptor system transformation, while"Atlantic" are not suitable for two kinds of transformation system.(5) The94antisense gene FtsZ1plants were obtained. Including38strains"Gannongshu No.2",27strains "Shepody",20strains "Favorita" and9strains "Atlantic".(6) Antisense FtsZ1gene express and suppress the chloroplast divisionof potato plants.
