In order to study on the antibacterial molecular mechanism of Aqueous Emulsion (AE) of Tagetes II, through mutagenesis to screening the mutant. Through transcriptome of high-throughput sequencing in the wild type and mutant, obtaining a lot of information as the foundation. I selected with significant difference in the expression, which are associated with the energy metabolism of pyruvate decarboxylase and glyceraldehyde-3-phosphate, and the p-tubulin regulated gene expression by the glyceraldehyde-3-phosphate, study on the gene expression and protein structure, hope to provide theory for further research to clarify the mechanism of molecular interaction of Aqueous Emulsion (AE) of Tagetes II.First of all, analyzed the RNA-Seq results, compared with the expression, Foldchange value, P value, as well as the three aspects of the reference from the protein, screening of β-tubulin, pyruvate decarboxylase, glyceraldehyde-3-phosphate dehydrogenase as the object of study. Second, according to the results of RNA-Seq, design the primer and cloning gene for the aimed protein. Depend on the gene sequence of β-tubulin, pyruvate decarboxylase and glyceraldehyde-3-phosphate by bioinformatics analysis, prediction and analysis of the three ORF protein, primary structure, spatial structure and partial nature.Firstly, depend on gene cloning, obtain the coding gene of β-tubulin, pyruvate decarboxylase and glyceraldehyde-3-phosphate.Secondly, three kinds of protein gene expression changes not due to gene mutation, is most likely the changed level of transcriptional regulation.Thirdly, though the bioinformatics study, β-tubulin and pyruvate decarboxylase and glyceraldehyde-3-phosphate, structure and function of three kinds of protein is matching exactly.Results show that, the influence of Aqueous Emulsion (AE) of Tagetes mechanism in many aspects, one of that maybe by the change of gene expression and play a rule of sterilization.
