This paper took three cultivation modes of plants, including whole plants, callusand suspension cells as study objects. The effects of two oligosaccharides withdifferent concentrations on biological activity of plants, and the effects of regulatingplants’ growth and eliciting plants’ defense responses of oligosaccharides werestudied.The seeds of maize were immersed in oligosaccharide solution with differentconcentrations, and the effects of two oligosaccharides on maize seeds germinationand seedling growth were studied. Four basic growth indexes: shoot’s height, root’slength, shoot’s weight and root’s weight and two biological activities: content ofchlorophyll, activity of root system were measured. The results showed that twooligosaccharide had no positive effects on germination rate of maize seeds, whilepromoted the growth of seedling of maize. The best promoting concentration of ADOwas100.0mg/L. When the concentration of COS was10.0mg/L, root’s length, root’sweight and shoot’s weight were higher than other treatment groups. But when theconcentration of COS was50.0mg/L, the shoot’s length was the higher than othertreatment groups.Tissue culture technique was used in the second part of this paper, and the effectsof oligosaccharide on formation of callus, regeneration of adventitious shoot andadventitious root were studied. The hypocotyl of tomato and the cotyledon ofnon-heading Chinese cabbage were used as explants. The results showed that in thecultivation of the hypocotyl of tomato, the best promoting concentration of ADO onformation of callus and regeneration of adventitious shoot was8.0mg/L. When theconcentration of ADO was1.0mg/L, it promoted the regeneration of adventitious rootof regenerated shoot effectively. All concentrations of COS inhibited the formation ofcallus and the regeneration of adventitious shoot while it promoted the regeneration ofadventitious root of regenerated shoot. The effective concentration of COS was0.1mg/L.The results showed in the cultivation of the cotyledon of non-heading Chinesecabbage, both ADO and COS promoted the formation of callus and the regenerationof regenerated shoot. The best concentration of ADO was1.0mg/L, while COS0.5mg/L. In addition, ADO could promote the regeneration of adventitious root ofregenerated shoot, while COS inhibited this process. The effective concentration ofCOS was0.5mg/L.The growth index and activity of defense enzyme of tobacco suspension cell wasresearched in the last part of this paper. The results showed that both ADO and COSpromoted the growth of suspension cell. The dense volume, dry weight and activity of defense enzyme of cell by adding different concentrations of oligosaccharide werehigher than which adding no oligosaccharide. The effective concentration of ADOwas50mg/L, while COS was10mg/L.
