Meiosis is a peculiar way of sexual reproducing organisms in the cell division, some fragments between non-sister chromatid of homologous chromosomes during meiosis exchanged, this phenomenon not only, increased the genetic diversity of genes, but provid more raw materials to natural selection. Meiosis is the generational turning point of sexual reproduction species, while homologous recombination is the key of meiosis.SPO11is a homologous protein of the Ⅱ topoisomerase, it can catalyze the formation of DNA double-strand breaks, thus starting the meiotic recombination. SPO11gene in Arabidopsis contains three homologous genes spo11-1,spo11-2and spo11-3. Compared with protein sequences of spo11in other species there is20%to30%similarity.Three genes play a role in two pathways, spo11-1and spo11-2genes play a major role in meiotic recombination and there are interaction between the two genes, spo11-3mainly plays a role in DNA repair process. These three are all meiosis-specific proteins, and also are necessary proteins in DSB formation process.Spo11are present in many species, which indicating that the protein is conserved. The main results are as follows:1、using the method of bioinformatics to analysis the gene of SPO11and its encoded proteinsuch as the open reading frame, encode the amino acid sequence,analysis the physical and chemical properties of genes, hydrophobic sites, transmembrane regions, signal peptides, and proteins structural sites we can understand more clearly about the regulatory mechanisms of meiotic recombination.2、Obtained the genes by electronic cloningAs the EST database of maize and oilseed rape is very rich, and very few studies on the spo11gene, so this study of electronic cloning selected in maize and oilseed rape database. Respectively Arabidopsis SPO11-1, SPO11-2, SPO11-3gene as a probe sequence, using the BLAST search tool to obtain some sequence similarity, use CAP contig assembly program, at alst obtain cloned sequences in maize and oilseed rape.3、Bioinformatic analysis of protein of SPO11in different species Using bioinformiatics methods to analysis these genes, such as conserved amino acid sites, conserved domains and protein superfamily and so on. Analysis shows: SPO11protein have five conserved regions, and four conserved amino acid in different species, indicate SPO11proteins in different species have a certain conservatism. In addition to corn protein SPO11-1protein all the SPO11hace domains TORPIM_TOPOIIB_SPO, except the three SPO11homologous genes of oilseed rape all contain TP6A domain. SPO11genes of three species belong to two different superfamily TORPIM and TP6A. The emergence of electronic cloning, makes the fact that clone a new gene in a shorter time become possible.
