The human amniotic mesenchymal stem cells (hAMSCs)has stem cell characteristics. Because of its easily obtained, extensive sources, and so on. The human amniotic mesenchymal stem cells has become the focus of national research in tissue engineering and cell replacement therapy. In recent years,the hAMSCs as a technical means alternative to cell transplantation in the treatment of various diseases, and it has better clinical results. In this paper, the author use the means of isolation and culture hAMSCs, and identify the biological characteristics, differentiation potential to explore and make experiment. Through the amniotic mesenchymal stem has contained cells (MSCs) in vitro separation, identification and induction, and to make sure whether it is stability and practicality, making the cells becomes the initial purpose of the trial and then to lay the foundation for the next animal experiments.In this experiment, to establish a stable inter-culture system of human amniotic mesenchymal cells, first obtain passage cells under sterile conditions. And then induced by different media to nerve tissue, inducing osteoblasts, adipocytes and induce cells to insulin.By the study of differentiation potential between the human amniotic mesenchymal stem cells,and mesenchymal morphology of human amniotic cells,and cell proliferation properties, cell viability assays and use immunohistochemical methods to identify them. The results show that:1By successfully established the system of cultured human amniotic mesenchymal cells, after primary cell culture, and continuous to culture the passage cells, and then find that from the six generations emergence of the differentiated cells,becoming old and in poor statuation gradually.The human cultured cells has been identified which is the amniotic mesenchymal cells, so we decided to choose the3-5passage cells as our test object.2We also has identified that the human amniotic mesenchymal cells with the stem cell characteristics. Though the Prompt identification results between human amniotic mesenchymal cells that expressed stem cell surface marker SSEA-4, the expression of stem cell pluripotency-related genes Oct-4. So,the experiments expression that the human amniotic membrane obtained the pluripotent of the mesenchymal cells.3The experiment result shows that by human amniotic mesenchymal cells in vitro differentiation potential. and the human amniotic mesenchymal cells can be induced to differentiate into mature nerve cells, generated mineralized nodules osteoblasts and adipocytes with lipid droplets and and double diphenylthiocarbazone stained insulin-like cells.Through the identification and induction it is confirmed that the cultured cells has a typical characteristic of mesenchymal cells and their growth is in good condition,besides it can be still used the next studies of experiment suitably. |
