Aerobic anoxygenic photosynthesis bacteria is under aerobic conditions of organic matter, sulfide or ammonia as hydrogen donor, by a bacteriochlorophyll capturing light for photosynthesis, but not a class of prokaryotes for release oxygen.At present, AAPB’s research focuses on the marine environment, Study of Classification and physiological and biochemical functions on freshwater lakes AAPB is very less.In this study, Inner Mongolia plateau lakes’surface water as the research object, Isolation and purification of Aerobic Anoxygenic Phototrophic Bacteria from the Inner Mongolia Plateau Lakes.Use the crossed coating methods.Then assay on denitrification activity and cloning of denirification gene. The results are as follows:(1)Cumulative isolated from15highland lakes samples get1246of bacteria, belonging to36genera. Including175for the AAPB, the proportion reached14%, belonging to21genera.(2) Among AAPBs, W2and W38strains have denitrification activity.Their total nitrogen (not sterilized) removal rates were36.48%and30.57%; ammonia nitrogen removal rate was95.66%and93.72%.(3)The test results of Denitrification gene function show that the strain of amoA positive was detected only two strains of the genus Sphingomonas; Detects the two strains of the genus Microbacterium strains may exist norB; There are10generas of18AAPB nosZ gene was detected bands; Belonging to12genera19AAPB detected nirK presence; has16genera of31AAPB detected nirS.The results of this study show that the Inner Mongolia Plateau lakes contains abundant AAPB groups, for the full understanding of the diversity and abundance of AAPB foundation in freshwater lakes. And part of this strains has a good denitrification, its function of protecting aquatic ecosystems play an important role.
