| F-box protein is involved in many biological processes in eukaryotes. F-boxprotein characterized by an F-box motif is a subunit of SCF complex, which works asdeterminant in substrate recognition. SCF complex is a very important ubiquitin E3ligase which has been exploited very well in plants. Currently, many F-box proteinshave been identified in plants which are involving in hormone signal transduction andbiological processes, such as self-incompatibility and floral development. F-boxproteins may also participate stress response in plants. By Promoter cis-actingelement analysis software prediction the promoter of FOA2gene(F-boxoverexpressed/oppressed ABA signaling2) which encodes a F-box proteinfunction,found there are many light response components. Therefore, this paperpreliminary studied light regulation expression model and function of the gene FOA2。The main research results made the following:Using real-time quantitative PCR analysis of the FOA2genes expression level inresponse to light, results showed that,mRNA and protein expression level of the geneis induced by blue, red and far-red light. And this study found that the mRNAexpression level of FOA2gene changed rhythmically in response to daylength.Analysis the FOA2gene tissue specific expression model found that it expreesedin every organs, but express higher in flowers and silique. Detected the expressionlevel of FOA2under silique maturation process,founde the expression level of FOA2Increase with the growth of mature silique of time. And Analysis the expression levelof FOA2under Seed imbibition process, founde the expression level of FOA2reducewith time of imbibed seeds.Statistics germination of FOA2overexpression transgenic and wild-type seedunder different light conditions,discovered that FOA2overexpression transgenic seedgermination rate is lower than that of wild-type,and FOA2overexpressing transgenicseeds under the far-red light conditions have more obvious different than wild-type.Treated seeds with different concentrations exogenous GA under white light, foundGA can restore FOA2overexpression transgenic seed lower germination ratephenotype. with different concentrations paco, ABA treatment, Overexpressingtransgenic seed germination is more sensitive to paco and ABA than wild-type,loss offunction mutants is more sensitive to paco and ABA than wild-type. Detected the expression level of some key genes in GA and ABA anabolicapproach in wild type and mutant by Q-PCR. The results showed that GA3ox2genespromoting germination expressed lower in FOA2than wild type and GA2ox2ã€ATNCED9and RGL2genes inhibiting germination expressed higher in FOA2thanwild type. There is an opposite result in foa2. These results illustrate that FOA2mayfunctions negatively in The GA synthesis pathways to regulate of seed dormancy.And FOA2may positively control ABA synthesis or signal transduction pathway toregulate seed germination.Yeast two-hybrid results display FOA2interact with ASK1, ASK2, ASK11,which are members of ASK gene family. So, presumably FOA2possible functions asan SCF complex component regulating seed dormancy and germination. |
