In the present study, five O. margaretae frogs were captured from the Guizhouprovince and a total of28cDNAs encoding17bioactive peptides belonging to14families were cloned from the skin cDNA library of O. margaretae. Amongthe17bioactive peptides, only one (Brevinin-1-Omar5) had beencharacterized. In order to understand their roles in host defensereactions, threebioactive peptides (Odorranain-H-OM1, Odorranain-M-OM andRanatuerin-2-OM), which possess low sequence similarity with the knownamphibian bioactive peptides, were selected for further chemical synthesis andfunctional analysis. Odorranain-H-OM1showed direct antimicrobial activityagainst bacteria and fungi. Odorranain-H-OM1showedanti-tumor activityagainstfive cell lines.Odorranain-M-OM exhibited concentration-dependentanti-oxidant activity. Ranatuerin-2-OM showed lectin-like activity and couldstrongly hemagglutinate human intact erythrocytes with or without thepresence of Ca2+. The diverse activities of bioactive peptides implied that theymay play different roles in host defense reactions of O. margaretae. |
