The mechanical property of cell has a relationship with the growth of celland the regulating function of itself. The correlation between deformabilityof cells and diseases is well-known, and has been evaluated in cell filtration andcell poking· experiments. The development of AFM make it possible to detect theproperty of single cell via the mechanical method. And AFM is also popular in theNanobiology on account of the high precision of AFM and can do the measurementin the biological environment etc. AFM can not only scan the sample at micro-nano size, make the things cannot be seen by eyes being seen through the way oftouching, but also do the nanoindentation experiment to explore the mechanicalproperty of the sample because of the load on the sample can be detected by thedeformation of cantilever.In this article we study the elastic modulus of living cells by AFM. Andexplore the experiment to measure the adhesion of living cells in biologicalenvironment, evaluate the practicability of this method. At last we establish aquery database that can query the mechanical property of cells.First, in this paper, we select the appropriate model for the measurement ofcell elastic modulus with AFM indentation, and analyze the experimentparameters of AFM on living cells indentation. Comparing with the previousexperiment methods, we present that the impact of indentation points and rate thatapproaching to living cells affect the elastic modulus of cells. And we designexperiments with a single variable method to verify the conjecture, and furtherimproving the elastic modulus with AFM measurements on living cells. Measuringthe elastic modulus in the cells of living cells, in less than10consecutive timesand the probe velocity1μm/s can make indentation modulus of elasticity of livingcells more reliable measurement results.Then,taking account of the adhesion of cells in human body is the adhesionability between cell and cells or cells and the substrate, and has relationship ofcommunication between cells. Using the AFM put the cell as a probe to measurethe adhesive force between cells (The SCFS experiment). Design the SCFSexperiment and evaluate the enforceability and the practicality of this experiment.We found that cycle of the SCFS experiment to detect the adhesive force of cells is long, and the condition of the experiment is instability, it is not suitable for alarge number of experiments.Last, we establish a small database management system that is in conventionof query and application.
