Abstract/Summary:
Oxysterol-binding protein related proteins are members of a protein family,including12members in mammals. They are called ORPs because of a commondomain that binds oxysterol. The investigations of mammalian ORPs show that ORPsplay roles in many aspects, such as transportation of sterols, regulation of lipidmetabolism, modulation of the actions of microtube-dependent lysosome andsignaling cascades and so on. ORP8,which is a member of ORPs family, has apredicted transmembrane fragment in the C-terminal. The target protein will locate atendoplasmic reticulum(ER) and extranuclear membrane through the fragment. Wehave researched releated function of ORP8and found that it involved in lipidmetabolism,however, the mechanism involved of ORP8remain unknown. Weconstructed ORP8recombined adenovirus vector and transfected into293A cells,successfully packaged the virus expressing ORP8. And then, detection by Westernblotand fluorescent confirm that ORP8expresses in293A cells. Finally, we collectedhigh-titer adenovirus by purification and concentration. ORP8silenced cellsdisplayed enhanced migration, altered subcellular distribution of an interaction partnerof ORP8, nucleoporin NUP62,increased expression in cytoplasm. ORP8was shownto compete with heat shock protein(Exo70) for interaction withNUP62,providing the mechanism of ORP8in cell migration.
