Objective:To development a sensitive amperometric immunosensor for myeloperoxidase(MPO) in human serum, and accordingly predict the risk in patients with ACS effectively in the 3 months and 2 years.Methods:1. Firstly, the glassy carbon electrode (GCE) modified with Nafion film self-assembly of gold nanoparticles (GNPs) and thionine (THI) layer-by-layer by covalent bonding and electrostatic adsorbing;2. Secondly, the antibody of MPO(anti-MPO) were absorbed on the surface of monolayer of GNPs.3. Finally, horseradish peroxidase (HRP) block the Non-specific adsorption site on the electrode , meanwhile, to amplify the signal of response current.Result:1. It has good response to MPO with height of peak current linearly dependent on MPO concentration in a range of 2.5-125μg/L. The detection limit was 1.425μg/L(estimated at a signal-to-noise ratio of 3)and the response time was less than 30 s.2. The variance of intra-batch and inter-batch were less than 2.94% and 4.15%, respectively.3. In addition, the performances of the immunosensor were studied in detail.4. Correlation analysis showed there was significant correlation between this immunosensor and ELISA(r=0.96, p>0.05).ConclusionThis amperometric immunosensor for determination of MPO is used repeatedly, in addition, of high sensitivity , high specificity , unnecessary lableing , simple operation for estimate the risk of ACS. Thus , it is suitable for detecting serum MPO in clinical laboratory.
