Phytohormone is a natural substance which can regulate the life course and the physiological activitied of plant. Phytonormone research is very significant to agriculture, horticulture and other related fields, and calls for a rapid, precise and convenient measurement procedure due to the limited of the traditional phytohormonal analysis.Use of chemo/biosensing techniques to detect the meaningful bio-markers for the accurate and rapid diagnosis of clinical diseases as well as for the field screening and mass monitoring of the epidemic diseases has been a novel, attractive and hot topic in the current medical studies.Aptamer have been widely researched and used now. Aptamer are isolated from random-sequence nucleic acid libraries by"in vitro selection". They are synthetic oligonucleotides with high binding affinity for a broad range of targets, including small molecules, proteins, or even whole cells.In this paper, the detail materials are summarized as follows:(1)Establishment of icELISA–kit for the determination of abscisic acid In chapter 2, we synthesized the ABA -BSA conjugate coating ELISA plates.A indirect competitive enzy-linked immunosorbent assay (icELISA)–kit was developed for the quantitative determination to ABA. The result showed ELISA-kit was simple , rapid, sensitive。The linear range of the assay was between 10 ng/mL ~ 1000 ng/mL,with the detect ion limit being1.4 ng/mL,which demonstrated the reliability and practical feasibility of these proposed agriculture practice.(2)Aptamer-based biosensor assay systemsIn chapter 3,a homogeneous, colorimetric system for adenosine detection has been developed based on the aggregation of gold nanoparticles induced by the adenosine binds specifically to the aptamer probe probe.Excellent response characteristics (for example, sensitivity, selectivity and linear response range) are obtained using either UV/vis spectrophotometer or the naked eye.In chapter 4, a method for the rapid detection of leukemia cells in whole blood samples has been developed based on a group of aptamers for the specific recognition of leukemia cells and the sensitive piezoelectric crystal sensor. The developed method is expected to hold great promise in medical diagnostics due to the ease of implementation, high sensitivity and specificity.
