Along with the development of medical imaging equipment, the union of function image and anatomy image is a developed trend, nevertheless, the potential of image registration lies in synthesis processing to apply the information which got from these image formation equipment, as to obtain new information which being helpful in the clinical diagnosis, and plays the vital role in the tumor pinpointing, in cancer's early diagnosis and treatment.This article was mainly to research the gradient information and mutual information matching method, and applied it in two-dimensional and three-dimensional medical image registration. We studied from interpolation method, multi parameter optimization and similar measure aspects and so on, selected the most suitable algorithm to apply it in the medicine image matching through many times test, obtained the satisfactory experimental result. Regarding to the two-dimensional medical image registration, we utilized the method combining gradient information and mutual information, has solved the question of partial extreme value without spatial information when pure use the mutual information; Then, extended this algorithm to the three-dimensional medical image registration, in order to enhance the speed of three-dimensional image registration, we introduced the DRR technology, and the result is satisfying.
