Microfluidic Chip is also named as Micro Total Analysis Systems, μ-TAS or Lab-on-a-chip. It was reminded by Manz and Widmer at the beginning of 90's in 20 centuries, which has roots in Micro Electro Mechanical System (MEMS), electronics, biochemistry and surface chemistry. The micro-channels, micro-chambers, micro-heaters and temperature sensors on the chip are fabricated by MEMS technology on the substrate of glass, silicon, polymer for sample analysis.Microfluidic chip technology is one of leading edge technologies of modern analysis science. The performance of every micro fluid device which constructs the whole microfluidic chip system is very significant, especially to optimization of the whole system. Although microfluidic chip is widely applied, the mechanism of microfluidic on this kind of chip is far from well known. The research on the liquid fluid characteristic of micro fluid device can provide instruction to the chip design and analysis of its function. Simulation to the design can effectively reduce the cost and improve efficiency. Meanwhile, it is an important way to study micro fluid. In this dissertation, study of fluid field on two kinds of chips is presented, including simulation of the liquid fluid field on the chips with simulation software CoventorWare and Femlab, experimental validation and optimization of chip design based on the conclusion.The topics of this dissertation are presented below:1. A chip based on electroeution principle was presented for the recovery of DNA fragments, which can make the isolation and collection of target DNA fragments possible in a single process by switching electrodes.2. Setup numerical model based on intensive study of two software packages (CoventorWare and Femlab) to improve the original chip design which has some unreasonable points found in the nucleic acid purification experiment. An optimization design was made for the channel structure and the distribution of the electric field in DNA recovery chip. Numerical simulation is done to evaluate the influence of these factors, and experiments were also performed to validate the simulation result. Both theoretical analysis...
