As an optical molecular imaging modality, bioluminescence tomography (BLT) has become a valuable tool for the noninvasive detection because of its significant advantages such as high sensitivity, low cost and convenient application. It reflects changes in molecular and cell level through accurate location of the light source. BLT technology research includes three main modules, reconstruction algorithm, and simulation tool, system development. And the imaging system development is core of the entire study which is the fundamental method verifying the accuracy of reconstruction algorithms and simulation result.In this paper, based on the introduction of the existing BLT prototype system, a detailed performance analysis and testing of the system were carried out firstly, to obtain the optimal data acquisition parameters for bioluminescence imaging. The inaccurate object image distance problem is solved by auto focus realization using the current system. Auto focus is based on the energy gradient definition evaluation function and optimization pole search algorithm. Ground on the optimal data acquisition parameters and system optimization, a real mouse implanted with luminescent source experiment was designed and completed. Pre-processing the collected experimental data, reconstruction the surface energy and light source location verify the reliability of the imaging acquisition parameters and reconstruction algorithm.
