[Objective] To research Human umbilical cord mesenchymal stem cells to repair radiation induced tissues injury to provide theoretical basis for related radiation disease.[Methods] 80 male SD mice were randomly divided into 4 groups. The rats were irradiated with 2 Gy rays and induced radiation injury except normal control group, setting up the model of radiation injury. We transplant 1ml umbilical cord mesenchymal suspension and Nacl in 12 hours and 24 hours after the radiotherapy. Tissues were detected on the 1,2,7 and 14 days respectively after radiation. The index of spleen were determined. By HE staining,the pathological changes of the spleen,liver, intestine were observed. Hepatocyte cell suspension were collected to determine the content of in superoxide dismutase(SOD) and malondisldehyde(MDA) by ehemocolofimetry. Then general condition of mice were observed and make records.[Results]Radiation control group compared with normal control group, activity decreasing, but administration of human umbilical cord blood mesenchymal stem cell become better the same with the normal control group. HE staining showed that : Radiation control group compared with normal control group, have a significantly difference according to Histological scoring criteria. Radiation control group compared with normal control group, the antioxidase activity of the SOD was decreased( F=41.89~182.96,t=10.79~22.03,P<0.01) and the content of MDA increased(F=158.441~632.751,t=12.13~44.22,P<0.01) significantly inside of cells.Therapy group compared with radiation control group, the antioxidase activity of the SOD was decreased(F=41.89~182.96,t=4.78~13.12,P<0.01) and the content of MDA increased(F=158.441~632.751,t=5.44~15.00,P<0.01) significantly inside of cells. Retherapy group compared with therapy group, the antioxidase activity of the SOD was increased(F=41.89~182.96,t=1.68~10.15,P<0.05) and the content of MDA decreased(F=41.89~182.96,t=10.83~26.02,P<0.05)significantly inside of cells. the index of the spleen. Compared with Radiation control group, administration of UCMSC could accelerate the restoration of recovery of all these indexes with significant difference.[Conclusion]Human umbilical cord blood mesenchymal stem cell can reduce tissues damage and repair regenerate tissues cell and effectively improve function.
