| Objective: To investigate the mechanism of Jiedu Huayu Decoction on the fluid dryness and toxin in SS model mice.Materials and methods: 60 C57 mice by weight points into six groups: normal group, model group, hydroxychloroquine group, dry toxic soup is low, medium and high dose group. Model group, Chinese medicine group and Western medicine group of mice with SD induced rat submandibular gland immunity, preparation of Sjogren syndrome mouse model. The preventive medication, regular daily administration and observe mice the glossiness of skin, hair removal, body weight, food intake, water, saliva secretion, and submandibular gland, thymus index, submandibular gland pathological changes were observed by HE staining and ELISA method to detect the mice submandibular gland and serum IL-6, IFN- expression.Result:1. SS mice in model group compared with normal group, the glossiness of skin loss, alopecia, hyperactivity, excited, easy irritability; reduced food intake, weight loss; water intake significantly increased; modeling started in the 2nd week, salivary flow in the model group compared with normal group decreased, but there was no statistical difference. Model third to the end of the experiment, The model group of salivary flow was significantly reduced(P < 0.05).2.The mouse submandibular gland index 2 model group significantly decreased, thymus index increased significantly. The between treatment groups were organ index between model group and normal group, which dry poison soup high dose group of submandibular gland and thymus index were the closest to the normal group.3. HE staining was observed in the submandibular gland pathology showed: stromal and glandular tube surrounded by a large number of lymphocyte infiltration; acinar atrophy, ranging in size, or even destroy; glandular atrophy, expansion, tube wall hyperplasia lesions. Proved successful. Compared with the model group, the treatment group was lighter than the model group. Among them, the high dose group was the lightest.4.The expression of IL-6 at low concentration of submandibular gland and serum of 5 normal mice. SS mice submandibular gland and serum IL-6 concentrations were significantly increased, which dry poison soup high dose group mice submandibular gland and serum IL-6 concentration closest to the normal group.5.The expression of low concentration of IFN- alpha submandibular gland and serum of 6 normal mice. SS mice submandibular gland and serum IFN- concentrations were significantly increased, which dry poison soup high dose group mice submandibular gland and serum concentration of IFN alpha closest to the normal group.Conclusion:1. Jiedu Huayu fluid dry poison decoction can effectively improve symptoms of SS mice, the increase in food intake, body weight, effectively alleviate the symptoms of dry mouth and reduce the amount of water, to increase saliva flow.2. Jiedu Huayu Shengjin method dry poison soup can reduce submandibular gland atrophy, increase the index of submandibular gland; reduce thymus index and reduce the occurrence of autoimmune.3 dry poison soup can effectively reduce the infiltration of lymphocytes, repair the inner wall of the cell and the inner wall of the cell, and inhibit the excessive self immune reaction.4. Dry poison soup can reduce of Pro inflammatory cytokines IL-6, IFN- content in serum and submandibular gland, which reduce the content of IL-6 in serum for the advantage; the role of IFN- than IL-6 weak.5. Inflammatory cytokines IL-6, IFN- in the serum and submandibular gland mainly invasion of submandibular gland mainly, and aggravate organism autoimmunity.6. Dry poison soup and hydroxychloroquine can effectively improve the symptoms of SS, dry poison soup curative effect is better than that of hydroxychloroquine curative effect and little side effect, safety. |
