Objective : By comparing different frequencies(2Hz and 100Hz) electricity for experimental observation of the rat sciatic nerve clamps damage,with the “Bcl-2”,“P53”,“Bax”as test index,reveals the curative treatment of sciatic nerve injury of antiapoptotic effect mechanism, seek the best of inhibiting neuronal apoptosis after peripheral nerve injury treatment frequency.Material and method:Reference clamps legal system as a model. Choose 48 SPF SD rats, weight of 250g-350 g, 7 to 9 weeks, male and female half and half. Using random Numbers table distribution method is divided into four groups: blank group, model group, treatment group 1(2Hz), treatment group 2(100Hz). HE staining Model group nerve fibers are arranged in the form of irregular, disorders, and around the nerve fibers of schwann cells increased, axons and myelin sheath cracking occurs, almost entirely disappeared. Treatment group pathological changes of nerve fibers is lighter, the emergence of new growth, nerve fibers both contrast treatment group 1 is lighter. 4. Western blot qualitative and quantitative detection 4.1 Western blot the Bcl-2 test results: Four groups of experiments the BCL-2 protein have obvious stripe, determination of grey value,(1) compared with the blank group, model group, treatment group 1, 2 BCL-2 protein expression in the treatment group increased significantly(P<0.01);(2) compared with model group, treatment group 1 and 2 the BCL-2 protein expression in the treatment group was obviously increased(P<0.01);(3) compared with the treatment group 1, group 2 BCL-2 protein expression in the lower(P < 0.05). 4.2 Western blot Bax test results: Four groups of Bax protein have obvious stripe in the experiments, the measurement of grey value,(1) compared with the blank group, model group and treatment group 2 Bax protein expression increased significantly(P<0.01), the treatment of Bax protein expression increased in group 1(P<0.05);(2) compared with model group, treatment group 1 and 2 Bax expression in treatment group was obviously lower(P<0.01);(3) compared with the treatment group 1, Bax expression increased in treatment group 2(P<0.05). 4.3 Western blot P53 test results: P53, four groups of experiments have obvious stripe, determination of grey value,(1) compared with the blank group, model group and treatment group 2 P53 protein expression increased significantly(P<0.01), P53 protein expression increased in treatment group 1(P<0.05);(2) compared with model group, treatment group 1 and 2 P53 protein expression inthe treatment group was obviously lower(P<0.01);(3) compared with the treatment group 1, P53 protein expression increased in treatment group 2(P<0.05). 5. Immune organized test results 5.1 The Bcl- 2 immune organized test results: Four groups of experiments the BCL-2 protein expression are,(1) compared with the blank group, the BCL-2 protein expression increased in model group(P<0.05), treatment group 1 and 2 the BCL-2 protein expression in the treatment group was obviously increased(P<0.01;(2) compared with model group, treatment group 1 the BCL-2 protein expression significantly increased(P<0.01), the BCL- 2 protein expression increased in treatment group 2(P<0.05);(3) compared with the treatment group 1, the BCL-2 protein expression increased in treatment group 2(P<0.05). 5.2 Bax immune organized test results: Bax protein expression are the four groups of experiments,(1) compared with the blank, Bax protein expression in the model group and treatment group 2 significantly increased(P<0.01), 1 Bax expression in treatment group were increased(P<0.05);(2) compared with model group, treatment group 1 and 2 Bax protein expression in the treatment group was obviously lower(P<0.01);(3) compared with the treatment group 1, Bax protein expression increased in treatment group 2(P<0.05) 5.3 the P53 immune organized test results: Four groups of P53 were expressed in the experiments,(1) compared with the blank group, model group and treatment group 2 P53 expression in increased significantly(P<0.01), P53 expression increased in treatment group 1(P<0.05);(2) compared with model group, treatment group 1 and 2 P53 expression in treatment group was obviously lower(P<0.01);(3) compared with the treatment group 1, P53 expression increased in treatment group 2(P<0.05).Conclusion: 1. EA can improve the acute injury of sciatic nerve of rat sciatic nerve functional recovery, and 2Hz group is better than that of 100 Hz.2. EA can be adjusted after acute injury of sciatic nerve clamped spinal motor neurons of anterior horn cells apoptosis activity and related proteins the Bcl-2, bax and the expression of p53, mechanism may be related to enhanced antiapoptotic effect factor to the Bcl-2, reduce apoptosis factors bax, the expression of p53. 3. Will adjust after acute injury of sciatic nerve clamped spinal motor neurons of anterior horn cells apoptosis activity and related proteins the Bcl-2, bax and the expression of p53, frequency of 2 Hz is better than 100 Hz.
