Expression And Function Of IL - 18BP In Hepatocellular Carcinoma

AimsThe aim of the study was to investigate the expression of IL-18BP in hepatocellular carcinoma and its relationship with the malignant phenotype of hepatocellular carcinoma.Methods1、Using mRNA detection, immunohistochemical(IHC) staining and further survival analysis of 117 cases of HCC with different levels of differentiation to detect the expression of IL18BP in cancer tissues and to determine its prognosis based on statistical data.2、In vitro the expression of IL-18BP in hepatocellular carcinoma cell line (97H) was interfered by siRNA, and its impact on the phenotype of the tumor cells was evaluated. Including clone formation assay, cycle arrest and apoptosis detection, and scratch test, EMT in cancer molecular marker detection.3、In vivo by interfering with the expression of IL-18BP, the change of tumor cells formation was observed while the detection about expression levels of cell cycle regulatory factor (Cyclin protein) are different.Results1、IL-18BP is highly expressed in human hepatocellular carcinoma, and it is affected by the factors such as tumor size, differentiation degree, portal vein tumor metastasis and other factors. The prognosis of patients with high expression is poor.2、In vitro, IL-18BP could promote the malignant phenotype of hepatocellular carcinoma, promote the proliferation of HCC cells, and inhibit the migration and proliferation of hepatocellular carcinoma cells after the knockdown.3、In vivo, the ability of tumor cells formation was found to be significantly weakened after interference of IL-18BP. The expression of CyclinDl, E decreased in the transplanted tumor constructed by shRNA.Conclusion1、 IL-18BP may be a novel cancer-promoting gene in HCC.2、IL-18BP may promote the malignant phenotype of hepatocellular carcinoma by acting in G1-S cycle of HCC cells.3、IL-18BP may be one of independent prognostic factors in HCC.
