Purpose:In this study, experimental method of serum pharmacology, molecular biology, as a stimulating factor angiotensinⅡinduced cultured rat mesangial cells EMC excessive deposition. Yu Tang Jiawei further observation on kidney mesangial cell proliferation film, and MMP-9 and TIMP-1 Inhibitor explore prevention mechanism for soup flavored jade kidney mesangial proliferative glomerulonephritis. Material and method:1. Apply the serum pharmacological method for preparing flavored jade serum containing kidney. 2. Angiotensin as an irritant to rat mesangial cells Ⅱwere cultured to proliferate. 3. Determination of rat mesangial cell proliferation by MTT assay. 4. ELISA assay matrix metalloproteinase-9 and tissue inhibitor of matrix metalloproteinase-1 content. Results: 1. Compared with the control group, model control group(Ang group) Number of GMCs Ⅱof rats significantly increased, the difference was significant(P <0.01, or P <0.05); Compared with the model control group, kidney soup flavored jade low, medium and high dose group and positive control group GMCs were significantly lower statistically significant(P <0.01, or P <0.05). 2. Compared with the control group, model control group MMP-9 were lower, the difference was significant(P <0.01, or P <0.05); compared with the model group, jade kidney soup flavored low, medium and high dose groups positive control group and MMP-9 content were all increased, the difference was statistically significant(P <0.01, or P <0.05); but the low kidney soup flavored jade, among the high-dose group had no significant dose-effect relationship. 3. Compared with the control group, model control group with higher levels of TIMP-1, the difference was significant(P <0.01, or P <0.05); compared with the model group, jade kidney soup flavored low, medium and high dose groups and positive control group TIMP-1 content are decreased, the difference was statistically significant(P <0.01, or P <0.05); but the low kidney soup flavored jade, among middle and high dose group had no significant dose-effect relationship. Conclusion: 1.Modified Jade serum containing kidney can upregulate the expression of matrix metalloproteinase-9. 2.Jade flavored kidney soup containing serum can inhibit the expression of tissue inhibitor of matrix metalloproteinase-1. 3.Modified jade kidney soup containing serum affect the expression of the two indicators, exerts its therapeutic effect on mesangial proliferative glomerulonephritis.
