Purpose:Breast cancer has become the most common malignant tumor in women cancer.The American cancer society released figures shows that the incidence of breast cancer in women has been as high as 29%, at the top of the list, the mortality rate is 15%, after lung cancer ranked second.The cause of its death is forming of distal metastases by tumor cells metastasizing through menstrual blood, lymphatic metastasis, planting transfer methods such as disseminated to the internal organs, bone, etc.Transformation of epithelial mesenchymal(EMT), refers to the epithelial cells by specific program into a mesenchymal phenotype cell biological processes. SBEM as a molecular marker of breast tissue specificity, has the close relation with micrometastasis of breast cancer. But there are not many studies about their correlation. The purpose of this study is to observe the SBEM genes influence on breast cancer cell migration, invasion ability, to explore whether SBEM through regulating the EMT promoting breast cancer metastasis.Material and method: 1.Building the SBEM gene overexpression plasmid and down-regulated expression plasmid. Establish the MCF- 7SBEM gene overexpression and gene down-regulated expression cell lines by the Virus transfection stable transfection method. 2.Observe the steady cell morphological changes. 3.Using Western blot method to detect SBEM gene protein expression in the Stable transfection cell lines. 4.Transwell invasion experiment of ability to detect breast cancer cells. 5.Western blot method to detect EMT related epithelial phenotype markers and interstitial phenotypic markers genes protein expression.Results: 1.The cell morphology doesnot observe the changes 2.Western blot method to detect the SBEM gene expression,the results shows that thegene overexpression and gene down-regulated expression cell lines are established successfully. 3.The results of Transwell chamber invasion assay showed: The invasion potential of cells was decreased after downregulation SBEM;The invasion potential of cells was increased after overexpression SBEM gene.. 4.Western blot testing EMT related marker gene protein expression did not see obvious difference, no statistical significance.Conclusion: The invasion potential of cells was decreased after downregulation SBEM;The invasion potential of cells was increased after overexpression SBEM. SBEM gene is not through EMT way to change the breast cancer cells invasion ability, the specific ways remains to be further research.
