ObjectiveCornel and Semen Ziziphi Spinosae are Chinese traditional medicines which have pharmacological action and good clinical effect, and play important roles in clinical treatment of modern application. Loganin and jujuboside A are the main efficacy components of them, as the medicinal materials quality control indicators and the main undertaker of play efficacy. This study synthesized the artificial antigens of loganin and jujuboside A, and identified their immunogenicities in several ways, then preparated anti-loganin and anti-jujuboside A polyclonal antibody.Method1. The artificial antigen of loganin is prepared by sodium periodate oxidization. The structure of artificial antigen is identified by TLC, UV, MALDI-TOF-MS and molecular exclusion chromatography.2. The artificial antigen of jujuboside A is prepared by sodium periodate oxidization. The structure of artificial antigen is identified by MALDI-TOF-MS and molecular exclusion chromatography.3. Balb/c mice were immunized with the prepared artificial antigen. To determined the concentrations of the coating antigen and serum antigen. The specificity of the s polyclonal antibody was detected by ic-ELISA respectively.Results1. Preparation of loganin successful artificial antigen by MALDI-TOF-MS detection to calculate the coupling ratio of about 5.2. Successfully getting the polyclonal antibody of loganin with high titer and good affinity. Under optimum conditions, a linear relationship in the range of 0.039~10.0 μg·mL-1 and the titer was up to 1:40000.3. Preparation of jujuboside A successful artificial antigen by MALDI-TOF-MS detection to calculate the coupling ratio of about 4.4. Successfully getting the polyclonal antibody of jujuboside A with high titer and good affinity. Under optimum conditions, a linear relationship in the range of 0.01-10000 ng-mL"1 and the titer was up to 1:4000.ConclusionThe artificial antigen of loganin and jujuboside A which is prepared by sodium periodate oxidization has good immunogenicity and produces a high-titre polyclonal antibody. Polyclonal antibody of loganin and jujuboside A has been prepared in turn. It created the conditions of the corresponding monoclonal antibodies and had an important scientific value for the quality control and the study of metabolism in the body.
